The increasing antibiotic resistance of bacteria is becoming a major health problem worldwide and alternative approaches to antibiotics are thus urgently needed. Gram-negative bacteria produce a dense network of polysaccharides (PS) at their outer membrane surface. These PS, including lipopolysaccharides, capsular polysaccharides, and exopolysaccharides, are important virulence factors and protective antigens. Our research group have become interested in developing synthetic mimics related to the main surface PS expressed by pathogenic Burkholderia species, which are potential bioterrorism threats and the causative agents of melioidosis and glanders, two neglected tropical diseases. Once covalently linked to a carrier protein, these oligosaccharides could act as glycoconjugate vaccines against Burkholderia-associated infections. Furthermore, some of the synthesized compounds could also be used as potential inhibitors of sugar-processing enzymes involved in the biosynthesis of Burkholderia PS. In the following presentation, the synthetic approaches towards these compounds will be described as well as their in vitro and in vivo biological evaluation.
